OptiPrepTM The ideal density gradient medium for the purification of plasma membrane and domains

چکیده
OptiPrepTM offers three gradient formats for the isolation of the plasma membrane (PM). Continuous or discontinuous gradients (1-3h) Continuous gradients (12-16h) Flotation through discontinuous gradients OptiPrepTM offers two approaches to the purification of lipid-rich micro domains. The first to be developed involves the isolation of caveolae from a sonicated total plasma membrane fraction. This strategy has also been used for the isolation of lipid rafts.

OptiPrepTM The ideal density gradient medium for isolation of macromolecules and lipoproteins
The flotation of the different classes of human plasma lipoproteins in a centrifugal field by the sequential elevation of the density of the plasma by dissolution of KBr or NaCl/KBr mixtures has been widely used over many years. The operation is routinely carried out in a fixed-angle rotor and takes at least three days. In spite of its cumbersome nature, the technique is still widely used today.

1. Comparison with other density gradient media Compared to CsCl and sucrose there are procedural advantages to the use of OptiPrepTM: ♦ OptiPrepTM is a sterile solution of 60% (w/v) iodixanol; it is simply diluted with saline to prepare sterile gradient solutions. It is the only gradient medium manufactured under strict FDA and EU cGMP compliance. ♦ CsCl and sucrose are both toxic to cells. ♦ ...

Actual endotoxin levels in each batch are usually measured at < 0.13 EU/ml. Unlike manufacturers of other density gradient media, Axis-Shield do not have to offer special, more expensive, low endotoxin-containing products; all Axis-Shield Density Gradient Media have always been, and continue to be, produced to the same high rigorous standards. For every batch produced, a Certificate of Analysis...
متن کاملOptimization of The Cell Aggregates Method for Isolation and Purification of Human Granulosa Cells from Follicular Fluid
Objective Aspirated ovarian follicular fluids (FF) contain luteal granulosa cells (LGC) and other contaminating cell types. Several strategies such as the antibody binding methods, the flask method, the cell strainer, and the positive selection of granulosa aggregates after density gradient centrifugation, were tested as LGCs purification methods. Each of these strategies has its own advantages...
